The latter might provide a more comprehensive, multimodal view on the disease with its interacting mechanisms and might be more suitable to reflect disease pathogenesis. This multiscale approachalso called integrative disease modeling can combine functional and structural neuroimaging techniques, cerebrospinal fluid sampling, and genomic data and analyzes their intercorrelations with computational algorithms .
Therefore, in the following sections, we describe existing computational models of different scales and outline how far they can be linked to the biological concepts presented before.

There has been a transformation in the part played by neuroimaging in Alzheimer disease research and practice in the last decades. Major deterrents to the widespread use of amyloid PET remain cost and availability. In contrast, MRI and FDG PET may give an indication of a frontotemporal or vascular pathology when an amyloid PET scan would be ambiguously negative in both cases. The threshold of sensitivity of amyloid PET has yet to be precisely determined, but it is clear that some level of amyloid deposition is histologically detectable prior to the in vivo signal becoming positive .
